題目: 未利用バイオマスのカスケード利用による食糧油脂及びマテリアル変換産業の創出 ~マテリアル変換としてのセルロースナノファイバーの研究動向~

 生物多様性をはじめ地球環境問題がクローズアップされる中、サステナブル社会構築に向けた新たな取り組みが各産業界で行われています。その一つに石油資源に代わって再生産可能なバイオマス素材の有効利用及び価値変換に関する研究があります。今回、私は油脂産業論文の執筆にあたり、油脂産業で廃棄されているバイオマス系材料に着目し、更にそれらをカスケード利用することによる新たな価値創造、市場創出について提案させていただきました。本セミナーでは、その油脂産業論文の内容に加え、企業研究者として私が進めてきたこれまでの研究内容、そして現在の研究進行中のセルロースナノファイバーの応用研究についてもご紹介させていただきたいと思います。セルロースナノファイバーとは木材の場合、幅が約4nm、長さは数マイクロメートルで近年世界的に注目されている新ナノ素材です。皆様の研究の一助になれば幸いです。

題目:Characterization of novel marine bacteria with wide spectrum quorum sensing inhibitory activity

The term quorum sensing was coined to describe the ability of bacteria to perceive and respond to population density for the collective regulation of gene expression, allowing social coordinated behaviors. Since bacterial communication plays a key role in the control of many physiological functions in bacteria, including the expression of virulence factors in plant and animal pathogenic bacteria, there is great interest in the design and implementation of systems to inactivate QS, a process generally known as quorum quenching, that could function as a new antipathogenic strategy for the control of bacterial diseases. A survey of quorum quenching activity among marine bacteria carried out in the laboratory of Marine Biotechnology and Aquaculture of the University of Santiago de Compostela, directed by Associate Prof. Ana Otero, revealed that this enzymatic activity is extremely frequent among marine isolates and also in metagenomic collections. Some of the isolates, presenting a wide-spectrum quorum quenching activity have been characterized and the capacity to control pathogenic bacteria tested in vitro and in vivo. In the case of strain 20J, a member of the marine genus Tenacibaculum, a new lactonase has been cloned and the capacity to improve survival of mollusk larvae cultures has been demonstrated.

解説:細菌のクオラムセンシングを阻害する新たな物質が海洋性細菌で発見されました。クオラムセンシングとは、細菌が自分と同種の菌の生育密度を感知して、それに応じて物質の産生をコントロールする機構のことです。クオラムセンシングの阻害機構の解明は、細菌感染症の予防や治療に役立つことが期待されています。

本セミナーの演者であるドイツのHagemann教授、Hess教授は、シアノバクテリアの環境応答に関して世界をリードする研究を進めている著名な研究者です。筑波で開催される日中韓学生フォーラムの講師として来日される折に、埼玉大学でも講演していただけることになりました。Hagemann教授はシアノバクテリアの塩ストレスに対する応答について、Hess教授は最近明らかになってきたnon-coding RNAがシアノバクテリアの環境応答に果たす役割について、分かりやすく話して下さいます。大変貴重な機会ですので、学部学生から教員の皆様まで是非ふるってご参加下さい。17時半からはセミナー室にて、両教授を囲む簡単なパーティーを開催予定です。

題目: Salt Adaptation of Cyanobacteria - from Genes to the Environment

Changing or high concentrations of inorganic salts (salt stress) represent one of the main environmental factors determining the occurrence of microorganisms in different habitats. Salt stress includes at least two primary stress components. High cellular amounts of inorganic ions, i.e. Na+ and Cl-, exhibit direct toxic effects on macromolecules leading to inhibition of cellular metabolism. In parallel, high salt concentrations in the surrounding soak water out of the cell, diminishing turgor pressure and abolishing the capability of growth. Therefore, all cells evolved strategies to adapt to changes in the external salt levels, which include the active export of inorganic ions minimizing toxic effects and the accumulation of non-charged, low molecular organic compounds (compatible solutes) keeping water inside the cell. These acclimation strategies were studied among cyanobacteria with defined laboratory strains of different salt tolerance levels during the last 30 years. These studies allowed defining genes and proteins characteristic for freshwater, moderate halotolerant (marine), and hypersaline strains. Now, about 70 cyanobacterial genomes are available in data bases, which can be searched for the occurrence of salt resistance genes predicting salt acclimation strategies of cyanobacterial strains without extensive cultivation studies. Moreover, this knowledge combined with advance methods such as metatranscriptomics will allow analyzing salt-acclimation of cyanobacteria in diverse environments.

題目: The Regulatory System of Cyanobacteria: An RNA Perspective

For their exploitation in biotechnology and the establishment of 3rd generation biofuel production in photosynthetic cyanobacteria it is important to understand their regulatory system. Moreover, the ability of cyanobacteria for adaptation to vastly different environmental conditions suggests the existence of sophisticated regulatory mechanisms. Regulatory proteins can be expected to interplay with various types of regulatory RNA molecules within the different signal transduction pathways and stress responses. However, comprehensive data on the architecture and composition of the cyanobacterial transcriptome have been lacking for a long time. More recently, we have experimentally characterized the transcriptomes of the unicellular model organism Synechocystis sp. PCC6803 and of the nitrogen-fixing filamentous Anabaena sp. PCC7120, identifying more than 3,000 and 10,000 active promoters, respectively [1-3].
Our results suggest a surprisingly complex transcriptome architecture: Only about 35% of all promoters drive the expression of protein-coding genes and operons, whereas the remaining 65% transcribe various types of non-coding RNAs. The functional characterization of this class of potentially regulatory RNA molecules is a constant challenge. We show some of these non-coding RNAs fulfill distinct roles in the control of stress responses [4,5] and the optimization of photosynthesis [6]. Modelling the molecular and functional interactions of regulatory RNAs globally suggests an array of distinct molecular mechanisms that need to be integrated into the fabric of the intracellular regulatory network. This work is of general interest for understanding mechanisms of gene expression control in prokaryotes and specifically relevant for the establishment of long-time producer strains of 3rd generation biofuels and for understanding the adaptation potential of photosynthetic primary producers in a changing world.

題目: 赤潮鞭毛藻の日周鉛直移動

多くの鞭毛藻類は、昼間は表層に、夜間は底層に遊泳して集積するという日周鉛直移動を行う。この能力により、鞭毛藻類は天候、透明度や成層構造などに左右されることなく光合成に必要な光を獲得できる、幅広い水深層の栄養塩を利用できるなど、移動能力に乏しい他の微細藻との増殖競合の上で、大きなアドバンテージを有していると考えられている。日周鉛直移動は微細藻類の生態を考える上で重要な生物現象であるが、現象の観察には多大な労力を要するため、鉛直移動に与える環境条件の影響や移動リズムを引き起こす細胞内のしくみについての知見は極めて限られている。 一方で、私どもは最近、実験室レベルで多数のサンプルを同時にかつ無人で観測できる日周鉛直移動自動観測システムを開発した。この技術革新により、赤潮原因ラフィド藻シャットネラが微弱な光を青色光受容体で感知し、光環境の変化に合わせて鉛直移動リズムを調節している可能性を見出した。

題目: 転写抑制による植物の発生制御 -- メリステムの機能と花の発生

植物の発生は、転写調節、シグナル伝達、細胞間コミュニケーションなど、さまざまな過程で制御を受けている。花の器官を決定する ABC モデルは、転写調節因子による制御としてよく知られている。ここ数年、転写抑制に関わる因子が、発生において重要なはたらきをしていることが明らかになりつつある。例えば、シロイヌナズナのTOPLESS遺伝子は transcriptional co-repressor をコードしており、そのドミナントネガティブ変異体は、胚発生において、茎頂分裂組織の位置から根が生じると言う劇的な表現型を示す。すなわち、根の発生を促進する一群の遺伝子を適切に抑制しておくことが、胚発生にとって非常に重要なのである。 私たちの研究室では、イネを対象に発生研究を行っている。最近、花の発生異常に関わる 2 つの遺伝子(ASP1とTOB1)を単離したが、いずれも、転写抑制に関わっていることが明らかとなった。本セミナーでは、この 2 つの遺伝子の機能について最新の知見を述べたい。

題目: Study of the structure of photosynthetic complexes and the effect of phosphatidylglycerol on cyanobacterial cell fission

We used differential scanning calorimetry (DSC) as a technique capable of identifying photosynthetic complexes on the basis of their calorimetric transitions. Annotation of thermal transitions was carried out with thylakoid membranes isolated from various photosynthetic mutants of Synechocystis sp. PCC 6803. The thylakoid membranes exhibited seven major DSC bands between 40 and 85ºC. The heat sorption curves were analyzed both by mathematical deconvolution of the overall endotherms and by a subsequent annealing procedure. The successive annealing procedure proved to be more reliable technique than mathematical deconvolution in assigning thermal transitions. The main DSC band, around 47ºC, resulting from the high enthalpy change that corresponds to non-interacting complex of PSII, was assigned using the PSI-less/apcE- mutant cells. Another band around 68–70ºC relates to the denaturation of PSII surrounded by other proteins of the photosynthetic complexes in wild type and PSI-less/apcE- cells. A further major transition found at 82–84ºC corresponds to the PSI core complex of wild type and PSII-deficient BE cells. Other transition bands between 50–67ºC and 65–75ºC are believed to relate to ATP synthase and cytochrome b6f, respectively. These thermal transitions were obtained with thylakoids isolated from PSI -/PSII-mutant cells. Some minor bands determined at 59ºC and 83–84ºC correspond to an unknown complex and NADH dehydrogenase, respectively. These annotations were done by PSI-less/apcE- and PSI-/PSII- mutants. Effect of phosphatidylglycerol (PG) deplition on cell division was studied by Synechococcus PCC 7942 and Synechocystis PCC 6803 pgsA mutant strains. The PG depletion resulted in an enlargement and elongation of cyanobacterial cells, which can correspond to formation of FtsZ ring. The ring formation could be blocked by elimination of PG from cell membranes. The formation of FtsZ ring was followed by GFP labeled protein in mutant cells. PG is crucial for the formation of division rings and is implicated in the fission of cyanobacteria.
